Orgovyx (relugolix) is an oral GnRH receptor antagonist used to treat advanced prostate cancer. It works by rapidly lowering testosterone levels, which prostate cancer cells need to grow, without the initial testosterone surge seen with some other hormone therapies.

How is this medication used?

Take a loading dose of three tablets (360mg) on the first day, then one 120mg tablet by mouth once daily at about the same time each day, with or without food. Do not stop taking Orgovyx without consulting your doctor, as testosterone levels will rise again.

What are the possible side effects?

Common side effects include hot flashes, fatigue, muscle and joint pain, constipation, and diarrhea. Hormone therapy can also affect bone density, mood, and sexual function, and may prolong the QT interval on heart tracings.

What are the potential interactions?

P-gp inhibitors such as erythromycin can raise Orgovyx levels (separate dosing may be advised), and combined P-gp/strong CYP3A inducers such as rifampicin can lower its effectiveness. Tell your doctor about all medications and supplements you take.
