Prograf, also known as tacrolimus, is a medication used to prevent rejection of organ transplants (e.g., kidney, liver, or heart) by weakening the body's defense system (immune system) to help your body accept the new organ as if it were your own.

How is this medication used?

Prograf is usually taken orally twice daily on an empty stomach or 2-3 hours after a meal. Dosage is based on your weight, medical condition, blood test results, and response to treatment. It is important to take it at the same times each day to maintain an even level of the medication in the blood.

What are the possible side effects?

Common side effects include shaking, headache, diarrhea, nausea, vomiting, upset stomach, constipation, trouble sleeping, numbness or tingling, or swelling of the hands/ankles/feet. More serious side effects can include high blood sugar, kidney problems, high potassium levels, and increased risk of infection.

What are the potential interactions?

Prograf can interact with a wide array of medications including, but not limited to, certain antibiotics, antifungals, calcium channel blockers, drugs used to treat seizures, and other medications that weaken the immune response. Grapefruit and grapefruit juice should also be avoided as they can increase tacrolimus levels in the blood to potentially dangerous levels. Always inform your healthcare provider of all the medications and supplements you are taking to avoid potential drug interactions.
